The aim of study was to do in vitro examination of spermatological characteristics in pure lines of cock semen, to determine spermatological parameters, to display differences between pure lines of Brown laying cock and to evaluate semen quality in Poultry Research Institute. In this study, in a total of 42 for 6 pure lines (Line 54, Barred Rock 1, Colombian Rock, Barred Rock 2, Rhode Island Red 2, Rhode Island Red 1), 7 cocks for each line, 35 weeks age brown laying cocks were used. During research, semen was collected from cocks by abdominal massage method in a total of six times, twice in a week throughout three weeks and semen was examined for spermatological characteristics. Averages of ejaculate volume (ml) sperm motility (%), sperm concentration (X10(9)/ml), the percentage of abnormal sperm (%), the percentage of dead sperm (%) and semen pH were recorded as 0.7 +/- 0.2, 0.9 +/- 0.2, 0.9 +/- 0.2, 0.6 +/- 0.2, 0.8 +/- 0.2, 0.7 +/- 0.1; 85.1 +/- 8.2, 87.6 +/- 7.7, 87.6 +/- 12.2, 87.6 +/- 7.0, 84.4 +/- 10.4, 85.0 +/- 9.0; 3.1 +/- 1.0, 3.6 +/- 1.0, 2.8 +/- 0.8, 3.3 +/- 1.2, 3.3 +/- 1.1, 3.2 +/- 0.7; 3.1 +/- 2.7, 3.0 +/- 2.1, 4.9 +/- 3.4, 4.5 +/- 3.5, 3.2 +/- 2.6, 4.8 +/- 4.3; 14.0 +/- 7.0, 13.5 +/- 5.9, 14.6 +/- 9.5, 12.0 +/- 3.9, 15.2 +/- 7.1, 14.7 +/- 6.4 and 7.0 +/- 0.3, 7.0 +/- 0.2, 7.1 +/- 0.3, 7.0 +/- 0.3, 6.9 +/- 0.3, 7.0 +/- 0.3 for Line 54, Barred Rock 1, Colombian Rock, Barred Rock 2, Rhode Island Red 2, Rhode Island Red 1, respectively. Differences among lines were significant for ejaculate volume (P < 0.001), sperm concentration (P < 0.01) and abnormal sperm (P < 0.01), but were found no significant (P > 0.05) for sperm motility, dead sperm and semen pH statistically. It is concluded that sperm characteristics in pure lines brown laying cocks is within normal limits and semen quality is good according to in vitro sperm characteristics in Poultry Research Institute by this study. |
