USING ITS2 SECONDARY STRUCTURE FORM TO EVALUATE GENETIC DIFFERENCES OF LATHYRUS L. GENUS FROM MIDDLE ANATOLIA OF TURKEY

Özet
In recent years, molecular regions of DNA were often used to clarify the genetic relationships in plants. Remarkably, ITS (Internal Transcribed Spacer-ITS1+ITS2) region was often used for molecular phylogenetic studies because of its highly repeated in number in plant genomes and large copy numbers that support PCR amplifications. Generally, ITS2 indicates particular differentiation among and between species; therefore, it is suggested as an important marker in molecular systematics studies. Although primary sequences of the region were useful, secondary structure of ITS2 region became valuable in species divergence and was used as a molecular morphological character in today. In the current study, representative Lathyrus L. species from Middle Anatolia of Turkey were chosen as samples. All studied sequences were retrieved from NCBI data bank for comparing genetic differences and sequences were aligned using ClustalW and Kimura-2 parameter to calculate the genetic distances. Both phylogenetic tree which was constructed with Maximum Likelihood method with Kimura-2 parameter additionally with G (gamma) distribution at MEGA X software and secondary structures of species which were predicted via the tools from the ITS2 database ( http://its2. bioapps. biozentrum. uni-wuerzburg. de/) and mFOLD web server were used for analyses. Based on the results, it was seen that both the overall genetic distances and the phylogenetic tree presented section separations of Lathyrus genus, and also ITS2 secondary structure predictions showed these genetic differences visually as a morphological character.
